Tries from Benji Marsh, Damian Tierney, Kellum Friday, Dan Rowett and two from Tom Bradfield did the damage with Rowett landing five from six conversions.
Chester started the game the stronger and quickly raced into a 10-0 lead with tries from Marsh and Tierney. Bradfield then crossed for his first of the game to put Gladiators 16-0 in front.
However, Westhoughton came back into the game and scored two tries in the final 15 minutes of the half to make the half time score 16-10 in Gladiators' favour.
Lions continued where they left off at the start of the second half and Chester then lost Craig Jones to the sin bin.
But some impressive defence kept the home side at bay and the Gladiators began to work their way back into the game.
Tries from Friday, Rowett and another from Bradfield pushed Chester well ahead on the scoreboard and ensured Gladiators ran out 34-10 winners.
Prop Chris Young was named as the man of the match.
