Gladiators had the perfect start when Declan Foy went over for an early try with Simon Quinn converting to put them six points up.
Quinn then limped off with a suspected torn groin and Warriors came back into the contest.
Leyland took full advantage of any mistake and took an 18-6 lead into the break after scoring three converted tries.
Chester continued to play some good rugby and crossed twice in the second period through Mark Thomas and Stuart Davies.
Leyland kept themselves ahead and punished some tired defence late on to make the scoreline lop-sided.
Dave Beddows, who came in for Quinn, was named as man of the match.
