Goals from Tendai Chitiza, Cosmos Matwasa, Harry Limb, Harry Vince and Tom Siddons secured an emphatic success in their first Northern Premier League South East Division match for 27 weeks.
They had last tasted victory in the league on March 14 when Spalding United were comfortably dispatched 5-1.
But the lengthy break due to the coronavirus pandemic had done nothing to take the sting out of Stamford's potent tail as they produced another ruthless display on their return to league action.
There was a minute silence before the match in memory of Daniels stalwart Michael 'Dutch' Holland who passed away on Friday.
An unchanged Stamford starting line-up from last weekend's FA Cup win over Diss Town were quickly on the offensive as they probed for an early opening.
Connor Bartle blasted a shot from distance wide while the visitors needed a couple of last-ditch blocks to keep the Daniels at bay.
It did only take 10 minutes to find their first breakthrough as Jordon Cooke's pinpoint crossfield pass picked out Chitiza who evaded a challenge before curling his shot home into the far corner of the Glossop net.
The Daniels continued to hold the upper hand as they looked to double their tally with the second goal arriving after 27 minutes.
Matwasa latched onto a lightning quick ball over the top and held off the challenge of Mambwe Chilufya before lofting a shot over the advancing Dale Latham in the Glossop goal.
The Stamford winger, with Chilufya in close contention, still had to make sure of the goal with a second touch home from close range.
Limb fizzed another shot over as Stamford went in search of a third goal while still keeping the door shut at the other end to deny Glossop a way back into the contest.
Stamford started the second half where they had left off as they looked to add a third goal to end Glossop's challenge.
Matwasa skilfully created an opening from Harry Vince's clever pass but Latham was down to save while the Daniels wideman and Cooke both had tentative shouts for a penalty turned down.
Skipper Sean Wright did well to thwart Leon Iluobe with a timely tackle as Glossop made a brief raid on the Stamford goal.
The game was then put beyond the visitors just past the hour mark when Limb latched onto Chitiza's swirling through ball and shrugged off the attentions of Devon Matthews before drilling an excellent finish past Latham.
The shackles were now well and truly off the home side who added a fourth goal just four minutes later.
Vince collected Chitiza's crossfield pass and burst past Chilufya who bundled him over in the box.
Vince - who had missed two penalties in the same game during pre-season - stepped up to confidently finish past Latham.
With one eye on Tuesday's home FA Cup clash with AFC Sudbury, Stamford started to ring the changes with three substitutions heading into the final 20 minutes.
But the Daniels still continued to dominate proceedings with Latham making a smart stop to keep out Cooke's header from a fine Rob Morgan cross.
Cooke then assisted his second and Stamford's fifth goal with 12 minutes remaining when his centre was met by an acrobatic overhead finish from Siddons.
Siddons and Dec O'Hare then both flashed late efforts wide as Stamford signalled their intentions for the campaign with an impressive opening salvo.
